Ataluren is designed to address genetic disorders caused by nonsense mutations, which introduce premature stop codons in the mRNA sequence, leading to truncated, nonfunctional proteins. By promoting ribosomal read-through of these premature stop codons, ataluren enables the production of full-length, functional proteins, thereby potentially ameliorating disease symptoms. This mechanism is particularly significant in treating Duchenne muscular dystrophy (DMD) resulting from nonsense mutations. DMD is a severe, progressive neuromuscular disorder characterized by muscle degeneration and weakness, affecting approximately 1 in 3,500 male births worldwide.
